PALM VITAMIN E, LIPID PEROXIDATION, HISTOLOGY, HISTOMORPHIC STUDIES IN RABBITS FED A CHOLESTEROL DIET Nafeeza MI1 , Norzana AG2 , Kamsiah J1 , Jalaluddin HK2 and MT Gapor3 Departments of Pharmacology1 & Anatomy2 , Faculty of Medicine, University Kebangsaan Malaysia and PORIM3 , Kuala Lumpur, Malaysia The "response-to-injury" theory states the mechanism involved on the genesis of atherosclerosis is endothelial cell injury. Injury can result from cytotoxic products from lipid peroxidation. In hypercholesterolemic states, the increase in oxygen free radicals intensifies lipid peroxidation processes which then leads to a change in endothelial cell membrane permeability. The leaky membrane then admits lipoproteins and other pro-atherogenic substances leading to atherosclerosis. Lipid peroxidation has been reported to be associated with hypercholesterolemia induced atherosclerosis. Therefore, antioxidants should then be able to reduce lipid peroxidation and suppress the development of atherosclerosis. To study the effects of palm vitamin E PV ; on lipid peroxidation and atherosclerosis in rabbits given a high a 2 % diet. Eighteen New Zealand White rabbits 2.0 2.8 kg ; were divided into 3 groups each n 6 the first group was fed a normal diet 100g day, the second group received a 2 % cholesterol diet 100g day and the third received a 2 % cholesterol diet 100g day plus daily oral administration of palm vitamin E at the dose of 60mg kg. After 10 weeks of feeding, the rabbits were killed and the aorta isolated for biochemical, histological and histomorphic studies. The aortic tissue cholesterol was done according to a method described by Sassen et. al 1990 ; . Tissue sections for histology was stained with H & E and Verhoeff van Gieson stains. Histomorphic studies were done on the sections stained with Verhoeff van Gieson using computerized image analyser, KS-400 Carl Zeiss, Germany ; . The aortic tissue content of total cholesterol were higher in groups that received a cholesterol diet compared to the normal diet group. There was no difference in the tissue cholesterol content in the groups that received a 2 % cholesterol diet with or without palm vitamin E. The palm vitamin E group had a lower content of aortic tissue malondialdehyde compared with the other two groups P 0.005 ; . There were no difference in the histological sections of the aorta in rabbits that received high cholesterol diet with or without palm vitamin E. There was also no difference in the thickness ratio of tunica intima to tunica media of aorta in both groups. Our findings suggest that despite a highly atherogenic diet, this dose of palm vitamin E had anti-lipid peroxidation effects. However, this effects seem to have no significant role in lowering the severity of atherosclerosis as indicated by various measurements of the inner layers of the aorta. Thus, although palm vitamin E at a dose of 60 mg kg was able to reduce the lipid peroxidation process but was unable to inhibit the progression of atherosclerosis. Sassen LMA et.al. Nitroimidazole; dimetridazole ; to its nitro radical anion was observed in agreement with the general scheme Fig. 1 ; . However, in the resistant strain, DMZ was reduced to its amine derivative R-NH2 ; through a low redox potential reaction. The addition of pyruvate, which can act as an electron donor through the PFOR complex Fig. 1 ; , increased the drug uptake 22. Hence, it was proposed that Nim proteins are 5-Ni reductases, which possibly use ferredoxin as the electron donor 22. Following the increasing availability of new bacterial genomes sequences, it emerges that Nim homologues are present in other genera of bacteria, including Deinococcus radiodurans, Bacteroides fragilis, Helicobacter hepaticus, Clostridium sp., Salmonella typhi, Streptomyces avermitilis, as well as in archea Methanosarcina sp. see Fig. 2 ; . Although the underlying physiological function of the Nim homologues in these organisms is unknown, it seems likely that the Nim gene family is ancient and widespread. More importantly, due to its unstable nature on mobilizable plasmids, Nim gene-based resistance poses a real threat to the existing applications of 5-Ni drugs. Indeed, a recent study identified seven Bacteroides resistant strains with minimum inhibitory concentrations of 32 g ml, all containing Nim genes 23. In this paper we present the first crystal structure of a Nim enzyme, the NimA from D. radiodurans drNimA, DR0842 ; at 1.6 resolution as well as its complexes with the MTR antibiotic, covalently bound pyruvate and covalently bound lactate. DrNimA shares 28% sequence identity and 54% sequence homology with bfNimA, and should thus be representative of the Nim family as a whole. Taken together with previous studies on NimA from B. fragilis and comparison with other closely related enzymes, our observations suggest that Nim proteins do indeed function as reductases. We propose a mechanism for the reduction of 5-Ni compounds by Nim enzymes that leads to generation of non-toxic derivatives and confers resistance against these antibiotics and advair.

Approved for use in rapidly progressive MS by the U.S. Food and Drug Administration FDA ; . Although it is clear that interferons act essentially on the inflammatory component of MS and not on the secondary degenerative process, this has not been clearly shown for mitoxantrone. The limitation in the use of this medication comes from its side effects, so much so that the AAN has not given it a level A recommendation. A lifetime dose limitation of 140 mg m2 is due to cardiotoxicity. Some 5% of treated patients complain of nausea, alopecia, UTI, amenorrhea, leucopenia, and elevated liver enzymes. Nausea, alopecia, leucopenia, and liver dysfunction respond well to cessation of the drug. A few patients, generally older, however, can remain amenorrheic. Five reports have been published of acute myeloblastic leukemia occurring between 3 months and 5 years after treatment. Although small in comparison to the total number of patients treated now approaching 20, 000 ; this is higher than spontaneous risk, and patients certainly need to be made aware of this deadly risk. Cardiotoxicity is a complication common to long-term anthracoid therapy. It appears to be dose related and has been shown to appear above 140 mg m2. Patients should be monitored for left ventricular ejection fraction LVEF ; by ultrasound or multiple gated acquisition MUGA ; scan and, if the LVEF is 50%, the drug should be stopped. Data reported by Ghalie and Edan on 2, 000 patients indicated no heart failure and, out of 12 patients whose LVEF dropped to 50%, only three did not recover. Our routine is to give mitoxantrone 20 mg m2 together with IV Solu-Medrol if no infection or leucopenia is present and if LVEF is 50%. We repeat this monthly for 3 months and, if the LVEF remains 50%, we repeat this quarterly for 18 months. One then gets close to the dose limit, the greatest drawback to the use of this medication. We are eagerly awaiting results of trials in which mitoxantrone and interferons are used sequentially, because it is probable that mitoxantrone will reinforce the effect of interferons. Mitoxantrone is an FDA-approved drug, and the use of mitoxantrone has spread. Vascularization. A. After instillation of two drops of 0.2 per cent Novesin Wander ; the central area of the cornea of each of six eyes of three adult albino rabbits Series I, Rabbits 1, 2, and 3 ; was cauterized using an electrically heated platinum wire, heated to red heat and applied to the cornea for approximately one second. The damaged area was then touched with solid silver nitrate crystals or stick ; so as to increase the tissue damage and render the ulcerated area more easily discernible. On the sixth day after cautery, the corneas were again touched with solid silver nitrate, as the limbal activity appeared to be subsiding. On the eighth day, all eyes were observed to have developed a hypopyon, and there was evidence of a purulent inflammation of the conjunctiva in four eyes. Treatment of these conditions was initiated immediately and took the following form: The cornea was anesthetized with 0.2 per cent Novesin and the whole of the cornea and conjunctiva was cleansed of exudate with a cottonwool swab moistened with an isotonic solution containing 2, 100 units of penicillin per milliliter. Powdered penicillin Sigma ; and Achromycin eye ointment Lederle ; were applied. This procedure was carried out twice on the eighth day and then once on the ninth and twelfth days. At this stage the conjunctival inflammation had subsided, the corneal stroma appeared nonedematous, the epithelium appeared continuous and intact, and the hypopyon was almost completely absorbed. During this period the corneas had become extensively vascularized. 73. Solid silver nitrate stick was applied to the center of the anesthetized cornea Novesin 0.2 per cent ; of each of the four eyes of two adult albino rabbits Series 2, Rabbits 4 and 5 ; for approximately two seconds. On the seventh day following the initial corneal damage, the same area of each cornea was cauterized for one second with a platinum wire heated electrically to a bright red color. Silver nitrate was again applied to the area for about one second. On the eighth day, a marked purulent corneal and conjunctival discharge was evident in each of the four eyes. Sulfacetamide ointment 10 per cent Evans ; was applied daily for three days, by which time the discharge had become nonpurulent. During this period all corneas had become extensively vascularized and amoxycillin.

Certain people are more susceptible or at greater risk of infection for a variety of factors. People who are very young or the very old are more at risk because their immune system may not be developed or may be waning. In addition, some medications, such as steroids and cytotoxic agents can increase infection risk. So can underlying diseases such as diabetes, blood disorders or cancer. Link 6 Microbes enter into the body Sometimes termed "portal of entry". In order for microbes to cause infection they must gain entry into the body. Different organisms have different ways of entering our bodies. For example, salmonella need to be ingested eaten ; . Some organisms may cause infection if they are inhaled e.g. tuberculosis. Others, such as hepatitis B, enter the bloodstream via broken skin, injection or sexual intercourse. Breaking the chain of infection Breaking the chain of infection by targeting one or more links can halt the spread of infection. This usually involves: a ; b ; c ; Eradicating the source of infection through appropriate antimicrobial therapy Preventing the method of spread through hand washing, hygiene, disposal of waste, decontamination of equipment etc or Protecting the individual at risk by immunisation Preventing microbes from entering the body by wearing protective clothing, using an aseptic technique when handling invasive devices, covering wounds and insertion sites with sterile dressings etc and acomplia.

Increasing the probability of congestive failure due to cq10 deficiency created from statins was not enough now we see that lowering cholesterol, the very thing that these cash cow drugs are literally designed to do can also cause congestive failure.

OPNAVINST 3120.32C CH-4 30 July 2001 630.17 EQUIPMENT TAG-OUT BILL a. PURPOSE. To establish fundamental requirements for a standardized and disciplined method of equipment isolation and de-energization. This bill establishes the Tag-out Users Manual NAVSEA S0400-AD-URM-010 TUM ; as the primary reference for establishing, maintaining, enforcing and training of tag-out program and tag-out processes used in all naval ships and craft. The Tag-out Users Manual provides: 1 ; A procedure, using standard tags and forms, to provide for personnel and ship safety, and prevent damage to equipment when a system, or portion of a system, is in an abnormal lineup or in an abnormal condition. 2 ; A procedure, using standard tags and forms, to provide temporary special instructions or to indicate that unusual action must be exercised to operate equipment. 3 ; A procedure, using standard labels, to identify installed instruments or gages that are unreliable or not in normal operating condition. b. RESPONSIBILITIES. [1] The Antiarrhythmics versus Implantable Defibrillators AVID ; Investigators. A comparison of antiarrhythmic-drug. Environmental concerns, such as the effects of aquaculture chemicals on water and sediment quality nutrient enrichment, loading with organic matter, etc. ; , natural aquatic communities toxicity, disturbance of community structure and resultant impacts on biodiversity ; , and effects on microorganisms alteration of microbial communities and the generation of drugresistant strains of bacteria ; . The general lack of knowledge concerning the effects and fates of chemicals and their residues in cultured organisms and within the aquaculture system itself. Similarly, information is lacking on the actions and fate of chemicals used in aquaculture in the aquatic environment in general impacts on non-cultured organisms, sediments and the water column ; . The lack of alternative means for chemical application. Development of highly specific targeted chemicals that have reduced side effects and environmental implicates is needed. The availability of affordable treatments suitable for aquaculture systems raising low-value species needs to be improved!

In future projects, Drs. Mandy O'Hara Murdoch University, Perth, Australia ; , a veterinary pathologist, and A. Bennett Jenson James Brown Cancer Center, University of Louisville ; will join us in fall 2006 to evaluate transgenic mouse models for papillomavirus oncogene-induced cancers in skin and hair follicles. Dr. Karin Jger Universittsklinik fr Dermatologie, Vienna, Austria ; will work on methods of analysis of hair fibers and nail plates, and Dr. Bruce Herron Genomics Institute, Wadsworth Center NYS DOH, Troy N.Y. ; will work on characterization of the repeated epilation Er ; mutant mouse. We periodically organize large international meetings on mouse pathology, and these have evolved to become a state-of-the-art conference, as demonstrated by the most recent meeting, held in Japan in 2003. A book was recently published as a follow-up to that meeting Sundberg JP and Ichiki T, Genetically Engineered Mice Handbook, Francis and Taylor, Boca Raton, Fla. 2005 ; . Several members of our group participated in the Australia-New Zealand Laboratory Animal Science Meeting in Perth, Australia in 2005 to bring this information to that continent. Current book projects include working with Drs. Matt Kaufman University of Edinburgh, Scotland ; and Alexander Nikitin Cornell University ; on embryology of the endocrine system. This project is nearly complete. We are also updating protocols on handling laboratory mice, performing detailed, systematic necropsies, taking gross and subgross photographs, and histopathology in preparation for the large workload associated with the Nathan Shock Center in Aging. As we assemble protocols we will convert this into a textbook.
