EVALUATION OF DIFFERENT STRATEGIES FOR THE REMOVAL OF TANNINS IN THE SCREENING FOR LEAD STRUCTURES Rack C., Matusch R. Therefore plants containing tannins often give positive results in screenings [1]. Since they are of little interest as lead molecules it is desirable to remove them beforehand. Precipitation methods are suitable for the removal of tannins in large number of samples which can be used for screening purposes. The aim of this study was the comparison of the effectivity of the precipitation of tannins by caffeine, lead acetate, gelatine and polyvinylpyrrolidine PVP ; . The influence of the pH between 4 to 10, the solvent methanol, ethanol and dimethyl sulfoxide DMSO and the amount of precipitation reagent was examined. A solution of tannic acid powder was precipitated with a solution of the precipitation reagent under the different conditions. After centrifugation the supernatant was mixed with a solution of ferric chloride. The iron-phenolate complex was spectrophotometrically determined [2]. At more acidic pH caffeine was the most effective reagent. At more alkaline pH lead acetate was most effective. At a pH above 7 no precipitation was observed using PVP. Parallel to an increase of the amount of solvent used the precipitation decreased. The effect was stronger for DMSO and less for methanol. Caffeine and gelatine were more influenced by higher concentrations of the solvent than PVP and lead acetate. The addition of increasing amounts of precipitation reagent showed that only lead acetate was suitable to remove the tannin completely. Our results indicate that the choice of the precipitation reagent for the removal of tannins should be tested individually for each specific screening condition.
